I've run into a new problem and I'm hoping that someone might be able to help me with a workaround for what appears to be a bug.
My ambulances refuse to spawn from the bay, and thus everyone who goes to the clinics or hospital are doing so using their own vehicles, and injured people are being left to die in their apartments. I've tried deleting the ambulance bays and replacing them, I've tried deleting the building and putting it back, both had no effect. The wellness vans from the hospital seem to go around the city just fine, as do fire, police, garbage, school, bus, recycling, and trade center vehicles.
Does anyone have any idea on how to resolve this? My city is surviving at 150k population and growing, but I think that my people would love to have some more help.

I have heard of this bug occurring to people across different services. I know one individual said he exited to region view and it corrected the issue. Other stated they simply had to leave the region and rejoin and that seemed to fix it.

Thanks! When I got back in one of the ambulances randomly was working, but none of the placed ones. Hopefully more will magically work in the morning.

I think this might happen when people tear down sections of their town with service vehicles on it. The vehicle just despawns and is still registered as an active agent in the game. The next time you have this issue, check how many vehicles are docked with the service building and whether it claims that they are in use or not.
I was not able to get around this problem when I had it. I just had to build enough vehicle bays to reach unused vehicle slots.

I now have 3 clinics in the city, all with at least couple of extra ambulances, and none of them are moving out despite the increasing number of sick people in my city.
I also noticed the school busses (about 4-5) of my high school are stuck in a small loop, unable to get anywhere.
This is happening to both of the cities I'm currently working hard on. Had to stop the simulation before further damage is caused, really need a hotfix for this.
I also tried restarting the game, exit to region, exit to main menu, nothing seems to be working.

I noticed the school bus loop once in the past. You can edit your high school and delete the school bus add-ons, then add new ones. It will fix that at least.
As for the idea that it's what happens when roads with service vehicles get destroyed, that makes total sense. This city is one that I was doing well on until a bug caused the nuke plant to melt down (2 cooling towers too), and then a number of disasters struck. I plowed everything down and started over with 4 million and a good amount of knowledge on how better to proceed with city building.

Has anyone figured out how to get destroyed ambulances to stop being unusable? I have to imagine that this could also happen in the event that a clinic, hospital, or the bays were destroyed by a disaster.

You have to go to the edit view of the clinic and then select the bulldozer button.

I've done that I'm afraid. Apparently the problem is that the buildings and the roads that the ambulances were on got bulldozed by me. As a result I can't get it to launch any ambulances unless I have enough garages to exceed the number of ambulances I had originally that are now trapped in some sort of ambulance limbo.

Anyone have any luck figuring this out? If I build like 3 or 4 medical centers and max out the ambulances I can get 1 or 2 that work but then your budget gets blown.

I'm afraid not. I made a new city and didn't run into the problem again, but my old city is still pretty much ruined as a result.
I also noticed that the new traffic efforts means that I can get a lot more coverage with a lot fewer services, so that's nice. Doesn't help with this, but it's nice to know.

Hopefully once they start moving from fixing server issues to gameplay issues this will be a priority fix. I have seen that other types of vehicles can disappear too. One type of service going down is definitely a hindrance but 2 or 3 would kill a city.

Hi guys.
I have the same problem.
I got an industrial high-tech city, and over time the wind shifted from south to southeast, engulfing half of my citizens in polluted air.
I deleted all clinics and built four hospitals, with some extra ambulances, but it seems like they just won't leave the hospitals at all, so all residents are actually walking to the nearest hospital.

Since this is clearly a bug, and a big one in my opinion, it needs to be fixed asap. Cannot really continue in my city with this problem. And I don't want to start over again just to have it happen again. Also, any software patch should fix existing cities too... not just new ones (!?!).

Did you also check out the education level of your population on the map? I wonder your school bus issues really have an effect or if the system still works despite the traffic issue.
